3. Alcohol Precipitation

• Precipitation: Add ethanol to the above concentrated solution under constant stirring until the final alcohol content reaches a specified volume fraction.
• Solid-Liquid Separation: Separate the precipitated solids from the supernatant using a tubular centrifuge or vacuum filtration equipment, and collect the supernatant rich in target components.

4. Cation Exchange Resin Purification

• Column Loading: Concentrate the supernatant from the alcohol precipitation step to recover ethanol, then dilute with water and adjust the pH to a slightly acidic level to ensure the target alkaloids exist in cationic form. Pass this solution slowly through a column packed with cation exchange resin. Alkaloids such as DNJ, which are weakly basic, are selectively adsorbed by the resin.
• Washing and Elution: First, rinse the resin column with purified water to remove water-soluble impurities. Then, elute with a specific concentration of ammonia water or alkaline ethanol solution, and collect the eluate with a high concentration of alkaloids.

5. Macroporous Adsorption Resin Refining

• Column Loading: Adjust the pH of the effluent or eluate from the cation exchange column to an appropriate level, then pass it slowly through a column packed with macroporous adsorption resin.
• Gradient Elution: Start by flushing the column with water or low-concentration ethanol to wash out water-soluble impurities, followed by elution with a higher concentration of ethanol to desorb the target components. Collect the target desorption solution.

6. Decolorization and Fine Filtration

• Decolorization: Add an appropriate amount of activated carbon powder to the collected refined solution and stir at a set temperature for a defined duration to remove dark-colored substances like chlorophyll.
• Fine Filtration: Use a plate-and-frame filter or titanium rod filter equipped with microporous membranes for precision filtration, ensuring complete removal of the decolorizing agent and producing a clear, transparent solution.

7. Concentration and Sterilization

• Vacuum Concentration: Transfer the decolorized clear solution to a vacuum concentrator, evaporate and recover ethanol at a low temperature, and concentrate to a paste form suitable for drying.
• Sterilization: Apply high-temperature short-time (HTST) sterilization or microporous membrane sterile filtration to the concentrated solution to control microbial limits.

8. Drying, Milling, and Sieving

• Drying: Feed the sterile concentrated solution into a spray dryer to obtain dry powder with low moisture content (<5%) in a single pass.
• Milling and Sieving: The dried particles may agglomerate; therefore, break them up using a high-efficiency mill and pass through a vibrating screen (80–100 mesh) to ensure uniform particle size distribution.

9. Final Blending and Packaging

• Final Blending: Transfer multiple batches of the sieved extract powder into a three-dimensional motion mixer and blend for sufficient time to ensure homogeneity and batch-to-batch consistency.
• Packaging: In a clean area, the blended powder is packaged into primary containers (aluminum foil pouches) using an automatic filling and sealing machine. Finally, proceed with outer packaging, labeling, and cartoning.
